How long should a 5000IU trigger shot stay in my system? I got a - preg test at 9DPO, then a + on 10 DPO?

I took several pregnancy tests at 10 DPO and all were positive. Now I have 12 DPO and had a beta version of blood test. The nurse called and said he was 44 is this too low?
